The invention relates to isolated mammalian nucleic acid molecules encoding receptor protein tyrosine kinases that are expressed in primitive hematopoietic cells but not in mature hematopoietic cells. Further disclosed are properties of the receptors encoded by such nucleic acid molecules, associated ligands for the receptors and the nucleic acid molecules themselves which encode the receptor protein tyrosine kinases such as murine flk-2, human flk-2, and murine flk-1. Also provided are methods for stimulating the proliferation and differentiation of primitive mammalian hematopoietic stem cells via contacting the stem cells with a ligand that binds to a receptor protein tyrosine kinase as expressed in primitive mammalian hematopoietic cells but not in mature hematopoietic cells. Therapeutic applications are included for the treatment of a variety of diseases.
