Summary:This invention describes a method for producing animal cells in chimeric or transgenic non-human animals which contain a desired gene sequence inserted into a predetermined gene sequence. The method permits the production of animal cells and non-human animals which have subtle and precise modifications of gene sequence and expression relative to natural non-human animals. After the DNA molecule with the specific, desired sequence is introduced into the mammalian cell, the introduced DNA molecule is then allowed to undergo homologous recombination with the predetermined gene sequence of the mammalian cell, thereby producing an intermediate cell; culturing of the intermediate cells, and the consequent intrachromosomal recombination are also disclosed.
