Described herein are methods by which antibodies specific for the cell surface markers Thy-1 and CD34 are applied to homogeneous stem cell compositions that have been obtained by the selective isolation of cells which are free of markers associated with differentiated cells but which display epitopic characteristics associated with the stem cells. Further disclosed are methods of maintaining the cells in an appropriate growth medium, and methods of assaying such stem cells for their capacity to generate members of each of the hematopoietic lineages. Therapeutic applications are included for the use of hematopoietic stem cells derived according to this method in the treatment of a variety of diseases. | | Abstract: | Human hematopoietic stem cells are identified according to expression of specific cell surface markers. Substantially homogenous stem cell compositions are obtained by selective isolation of cells free of markers associated with differentiated cells, while displaying epitopic characteristics associated with the stem cells. The stem cells may then be maintained by regeneration in an appropriate growth medium.
