Stem Cell Related Patent Number US5668104
Title: | Hematopoietic stem cell growth-promoting compositions containing a fibroblast-derived fragment of fibronectin and a growth factor, and methods employing them in vitro or in vivo | Inventors: | Nakahata, Tatsutoshi; Matsumoto, Japan
Kawano, Genji; Kamakura, Japan
Sudo, Tetsuo; Kamakura, Japan
Kojima, Katsuaki; Yokohama, Japan | Summary: | Described herein is a therapeutic composition comprised of a novel protein which promotes the growth of undifferentiated hemopoietic stem cells. The invention relates to the 18 kDa protein, denominated FDF-3, which has been isolated from cultures of human fibroblasts and has been found to potentiate the growth- and differentiation-promoting activities of certain cytokines toward hematopoietic progenitor cells. Amino acid sequences of the FDF-3 protein are disclosed, wherein one sequence corresponds to that of a C-terminal domain of human fibronectin. Therapeutic applications are included for use of the protein in combination with IL-3, G-CSF, GM-CSF, or stem cell factor (SCF) for the expansion of hematopoietic cell populations in vitro or in treatments which promote recovery from leukopenia or myelosuppresion in vivo. | Abstract: | An 18 kDa protein, denominated FDF-3, has been isolated from cultures of human fibroblasts and has been found to potentiate the growth- and differentiation-promoting activities of certain cytokines toward hematopoietic progenitor cells. The amino acid sequence of the FDF-3 protein, shown in SEQ ID NO: 14, corresponds to the sequence of a C-terminal domain of human fibronectin. The protein is useful in combination with IL-3, G-CSF, GM-CSF, or SCF (stem cell factor) in the expansion of hematopoietic cell populations in vitro or in therapeutic methods to promote recovery from leukopenia or myelosuppresion in vivo. | US Patent Website: | Click Here for Full Text of Patent | Title Number: | US5668104 | Application Number: | US1994000142449 | Date Filed: | 26/04/1994 | Date Published: | 16/09/1997 | Assignee: | Toray Industries, Inc., Tokyo, Japan |
