Farson, Deborah A.; Oakland, CA, USA | Summary: | Described herein is a method of transducing mammalian cells with a supernatant of cells transfected with retroviral packaging plasmids. The invention relates to a novel retroviral packaging system in which retroviral packaging constructs and packagable vector transcripts are produced from high expression plasmids by transfection in human cells, such that high titers of recombinant retrovirus are produced in the infected cells. Further disclosed are methods by which the novel retroviral constructs transduce primary human cells, including T cells and human hematopoietic stem cells, with foreign genes by cocultivation. Claims of the invention include the rapid production of high viral supernatants, and the efficient transduction of cells that are refractory to transduction by conventional means. This invention represents a continuation of other applications filed by the same assignee and inventors. | Abstract: | The invention provides a novel retroviral packaging system, in which retroviral packaging constructs and packagable vector transcripts are produced from high expression plasmids by transfection in human cells. High titers of recombinant retrovirus are produced in infected cells. The methods of the invention include the use of the novel retroviral constructs to transduce primary human cells, including T cells and human hematopoietic stem cells, with foreign genes by cocultivation at high efficiencies. The invention is useful for the rapid production of high viral supernatants, and to transduce with high efficiency cells that are refractory to transduction by conventional means. | US Patent Website: | Click Here for Full Text of Patent | Title Number: | US5686279 | Application Number: | US1994000258152 | Date Filed: | 10/06/1994 | Date Published: | 11/11/1997 | Assignee: | Cell Genesys, Inc., Foster City, CA, USA |
