The invention relates to a DNA construct comprising a disrupted mouse NPY Y1 receptor gene, wherein the disruption is the result of the insertion of a heterologous marker sequence into the gene such that the protein encoded by the disrupted mouse NPY Y1 receptor gene does not bind to NPY. Further disclosed are transgenic nonhuman mammals which have been engineered by homologous recombination to be deficient in the expression of the specific neuropeptide receptor. Also provided are the DNA constructs and engineered embryonic stem cells employed to produce such mammals, and assays which utilize either the animals themselves or tissue derived therefrom.
