Summary:This invention describes methods of generating the lambda (lambd) or P1 bacteriophage vectors that are useful in the targeted mutagenesis of eukaryotic cells and in the expression of genes and proteins. Also disclosed are methods for the identification of a lambd or P1 bacteriophage vector having a desired nucleic acid chosen from an assortment or library of bacteriophages, each of which possess a different nucleic acid insert. Further provided are methods for using such vectors in gene targeting and in the expression of genes and protein, as well as the precise conditions and procedures for growth, recombination and identification of the bacteriophage.
Abstract:The present invention relates to methods for the generation of lambda (.lambda.) or P1 bacteriophage vectors useful in targeted mutagenesis of eukaryotic cells and the expression of genes and proteins, methods for the identification of a .lambda. or P1 bacteriophage vector having a desired nucleic acid from an assortment or library of bacteriophage each having a different nucleic acid insert and the use of such vectors in gene targeting and the expression of genes and protein.
