Summary:Described herein is a method for stably transferring DNA into multipotent hematopoietic stem cells, thereby transducing extremely primitive hematopoietic stem cells with high efficiency. Further disclosed is the use of an adeno-associated vector for such transduction, said vector being capable of stably transforming highly primitive CD34+++CD38= cells which reside in a quiescent state, thereby retaining to a larger extent the ability to repopulate the hematopoietic system. Methods of DNA integration are provided, as are therapeutic applications in gene therapy for inherited metabolic diseases, viral infections, and cancer.
Abstract:This invention relates to a method for transducing extremely primitive hematopoietic stem cells with high efficiency, using an adeno-associated vector. This vector stably transforms highly primitive CD34+++CD38= cells which reside in a quiescent state and retain to a larger extent the ability to repopulate the hematopoietic system.
