Summary:This invention describes a method of creating a nucleotide construct which contains a positive selection marker flanked by restriction sites and sequences that can be treated to produce single stranded regions and which is useful for introducing targeted mutations into embryonic stem cells. The invention provides novel nucleotide constructs and the methods by which DNA constructs are made, such that sequences are introduced into the function of a gene in a cell, particularly in an embryonic stem cell, for purposes of disrupting that function. Therapeutic applications are included.
