Title:Methods of controlling proliferation and differentiation of stem and progenitor cells

Summary:This invention describes methods for expanding a population of stem and progenitor cells, in vivo or ex vivo, while inhibiting the differentiation of these cells, via a system of reducing the capacity of the cells to utilize copper by use of a transition metal chelator. Fully disclosed are the techniques by which the simultaneous expansion of a population of cells and the inhibition of the differentiation of the cells is performed. Included are methods for providing the cells with optimum conditions for cell proliferation while, at the same time, reducing the capacity of the cells to utilize cooper. The method has been successfully executed both in-vivo and ex-vivo. Central to this invention is the use of a transition metal chelator, which is provided to the cells and which binds to copper. The expanded, undifferentiated hematopoietic cells thus have a reduced intracellular copper content. Further disclosed are methods for obtaining the hematopoietic cells from a donor, growing them under the conditions described above, and transplanting the cells to a patient.
Abstract:A method of expanding a population of cells, while at the same time inhibiting differentiation of the cells, the method includes the step of providing the cells with conditions for cell proliferation and, at the same time, for reducing a capacity of the cells in utilizing cooper. The method can be executed both in-vivo and ex-vivo. A method of inducing differentiation in a population of cells, the method includes the step of providing the cells with a transition metal chelator which binds copper and which is effective in inducing cell differentiation.
